Toll样受体(TLR)信号通路PCR芯片可以同时检测84个在TLR介导的信号转导和先天免疫中发挥关键作用的基因。TLR家族的模式识别受体(PRRs)能够通过病原体关联的分子模式检测细菌,病毒,真菌和寄生虫。每个受体结合特异性的配体,进而起始针对特定病原体的先天免疫反应,并激活获得性免疫反应。例如,TLR4能够识别细菌脂多糖(LPS)或者是在血源性感染中引起感染性休克的内毒素。这些受体通过单体或异质二聚体的形式与受体蛋白相互作用,而从激发MyD88或TICAM1(TRIF)依赖的免疫反应。这些反应主要是通过NFκB分子激发信号级联反应。NFκB能够激活下游的JNK/p38信号或细胞因子分泌。这些信号通路的失活会导致严重的后果,并且引起多种自身免疫性疾病和病理性的炎症。这款芯片包含了TLR信号分子家族、受体、效应分子、以及受TLR激活的下游信号通路中的关键基因。这些下游信号通路包括:NFκB、JNK/p38、IRF 和JAK/STAT信号通路。通过实时定量PCR的方法,研究者即能够利用该芯片简单可靠地同时检测TLR介导的信号转导相关基因的表达。
